DNA topoisomerases from rat liver: physiological variations.
Besides the nicking-closing (topoisomerase I) activity, an ATP-dependent DNA topoisomerase is present in rat liver nuclei. The enzyme, partially purified, is able to catenate in vitro closed DNA circles in a magnesium-dependent, ATP-dependent, histone H1-dependent reaction, and to decatenate in vitro kinetoplast DNA networks to yield free minicircles in a magnesium-dependent and ATP-dependent r...
متن کاملGlucocorticoid-binding proteins of rat liver cytosol. I. Separation and identification of the binding proteins.
The specific interaction of glucocorticoid hormones with liver cytosol proteins of adrenalectomized rats was studied by an adsorbent technique for the separation of free and proteinbound steroid. By this rapid procedure a high affinity binding site for glucocorticoids can be detected which is usually overlooked when the slower technique of equilibrium dialysis is used. This metastable binding p...
متن کاملGTP-binding proteins in rat liver nuclear envelopes.
Nuclear transport as well as reassembly of the nuclear envelope (NE) after completion of mitosis are processes that have been shown to require GTP and ATP. To study the presence and localization of GTP-binding proteins in the NE, we have combined complementary techniques of [alpha-32P]GTP binding to Western-blotted proteins and UV crosslinking of [alpha-32P]GTP with well-established procedures ...
متن کاملBinding of chemical carcinogens to nuclear proteins of rat liver.
liver nuclei was studied. After the injection of the radioactive carcinogens into rats, liver histone fractions Fl, F2al, F2a2, F2b, and F3 were isolated by selective acid extraction. DNA and acidic proteins were extracted with 4 M CsCl at pH 11.6 and 14 and were separated by equilibrium density centrifugation. Histones and acidic proteins were purified by Sephadex chiomatography. Maximum level...
متن کاملBINDING OF NAPROXEN TO SERUM PROTEINS IN PATIENTS WITH LIVER CIRRHOSIS AND HEALTHY INDIVIDUALS
A three-fold decrease of the in vitro ability of serum proteins to bind naproxen was shown in patients with cirrhosis of the liver compared to healthy individuals. This decrease was caused by quantitative changes in serum proteins in the sera of patients with liver damage. Lower doses of naproxen are therefore suggested in the treatment of patients with liver dysfunction.
